UV-VIS Spectrophotometer and Its Supporting Instruments

The UV-VIS spectrophotometer is one of the essential instruments in laboratories, used to measure the absorbance and transmittance of a substance within the ultraviolet (UV) to visible light (VIS) wavelength range. To ensure optimal performance, various supporting instruments assist in the preparation, operation, and maintenance of this equipment, including:

  • Cuvette: A small container where liquid samples are placed, made of glass, quartz, or plastic, depending on the type of light used.
  • Light Source: The spectrophotometer utilizes different light sources, such as Tungsten-Halogen lamps for visible light (400-700 nm), Deuterium lamps for ultraviolet (UV) light (190-400 nm), and Xenon lamps for a broader spectrum covering both UV and VIS.
  • Monochromator and Optical Filters: These components help select specific wavelengths of light, separate them, and regulate light intensity for accurate analysis.
  • Light Detector: After passing through the sample, detectors such as Photomultiplier Tubes (PMT), Photodiode Arrays (PDA), or Charge-Coupled Devices (CCD) measure how much light is absorbed or transmitted by the sample. Blank and standard solutions are used as references to maintain measurement accuracy.
  • Software and Computer Integration: Modern spectrophotometers come equipped with software and computer connectivity, aiding in data recording, color analysis, and sample content evaluation, ensuring faster and more user-friendly results.

How a Spectrophotometer Works

  1. Light Source – Generates light at various wavelengths using Tungsten-Halogen (VIS) or Deuterium (UV) lamps as primary sources.
  2. Wavelength Selection (Monochromator) – The light is directed to a monochromator (prism or grating) to isolate specific wavelengths for analysis.
  3. Sample in Cuvette – The selected light passes through a cuvette containing the sample, where some light is absorbed while the rest is transmitted, depending on the sample’s concentration.
  4. Light Detection – The transmitted light is measured by a detector such as a Photomultiplier Tube (PMT) or Photodiode Array (PDA), converting it into an electrical signal.
  5. Data Processing – The electrical signal is converted into absorbance or transmittance values, which are then displayed on the spectrophotometer screen or transferred to a computer for further analysis.

UV-VIS Spectrophotometer at the Indonesian Molecular Institute (IMI)

The Indonesian Molecular Institute (IMI) utilizes a HORDE Electric UV-VIS Spectrophotometer, which enables direct measurements with a wavelength range of 190 nm to 1100 nm and a high resolution of 0.5 nm.
